Once you upload a level, it gets marked as uploaded (with a small up arrow icon in the level selection menu), and cannot be reuploaded unless you delete the uploaded level from course world.
The courses downloaded from Course World by pressing the download button in its course data will appear here, so players can play those course even without Wi-Fi access.
Super Mario Maker 2 does not allow you to edit downloaded maps. Whether this was something on Nintendo's mind, based on what happened in the previous game, is up for debate. In the main menu, scroll down and select Course World, which is the second to last option in the main list, just above Coursebot.
